Cognition Can Be Boosted in Some Patients With Down Syndrome, Study Finds – ScienceAlert

A new hormone treatment improved the cognitive function of six men with Down Syndrome by 10-30 percent, scientists said Thursday, adding the "promising" results may raise hopes of improving patients' quality of life.

However the scientists emphasized the small study did not point towards a cure for the cognitive disorders of people with Down Syndrome and that far more research is needed.

"The experiment is very satisfactory, even if we remain cautious," said Nelly Pitteloud of Switzerland's Lausanne University Hospital and co-author of a new study in the journal Science.

Down Syndrome is the most common genetic form of intellectual disability, occurring in around one in 1,000 people, according to the World Health Organization.

Yet previous research has failed to significantly improve cognition when applied to people with the condition, which is why the latest findings are "particularly important", the study said.

Recent discoveries have suggested that how the gonadotropin-releasing hormone (GnRH) is produced in the brain can affect cognitive functioning such as memory, language, and learning.

GnRH hormones regulate how much testosterone and estrogen is produced and increased levels of it help spur puberty.

"We wondered if this hormone could play any role in establishing the symptoms of people with Down Syndrome," said Vincent Prevot, study co-author and head of neuroscience research at France's INSERM institute.

The team first established that five strands of microRNA regulating the production of GnRH were dysfunctional in mice specifically engineered for Down Syndrome research.

They then demonstrated that cognitive deficiencies as well as loss of smell, a common symptom of Down Syndrome were linked to dysfunctioning GnRH secretion in the mice.

The team then gave the mice a GnRH medication used to treat low testosterone and delayed puberty in humans, finding that it restored some cognitive function and sense of smell.

A pilot study was conducted in Switzerland involving seven men with Down Syndrome aged 20 to 50.

They each received the treatment through their arm every two hours over a period of six months, with the drug delivered in pulses to mimic the hormone's frequency in people without Down Syndrome.

Cognition and smell tests were carried out during the treatment, as were MRI scans.

Six of the seven men showed improvement in cognition with no significant side effects however none showed a change in their sense of smell.

"We have seen an improvement of between 10-30 percent in cognitive functions, in particular with visuospatial function, three-dimensional representation, understanding of instructions as well as attention," Pitteloud said.

The patients were asked to draw a simple 3D bed at several stages throughout the therapy. Many struggled at the beginning but by the end the efforts were noticeably better.

The authors acknowledged some limitations of the study, including its size and that the choice of patients was "pushed by their parents".

"The clinical trial only focused on seven male patients we still have a lot of work to do to prove the effectiveness of GnRH treatment for Down Syndrome," Pitteloud said.

A larger study involving a placebo and 50 to 60 patients, a third of them women, is expected to begin in the coming months.

"We are not going to cure the cognitive disorders of people with Down Syndrome, but the improvement seen in our results already seems fundamental enough to hope to improve their quality of life," Pitteloud said.

Fabian Fernandez, an expert in cognition and Down Syndrome at the University of Arizona who was not involved in the research, hailed the "tour de force study".

He told AFP that while it is "difficult to envision" how such an intensive treatment could be used for young people, it might be better suited to delay the Alzheimer's disease-related dementia suffered by many adults with Down Syndrome.

It was also difficult to predict how such an improvement could impact the lives of people with the condition, he said.

"For some, it could be significant, however, as it would enable them to be more independent with daily living activities such as maintaining and enjoying hobbies, finding belongings, using appliances in the home, and traveling alone."

Male Contraception Pills Show Promise And They Have No Major Side Effects – SciTechDaily

Currently, male contraception is limited to vasectomies and condoms. The researchers hope to provide more options in the future.

A new study presented at ENDO 2022, the annual meeting of the Endocrine Society in Atlanta, Georgia, suggests that two experimental male contraceptive pills appear to successfully reduce testosterone without causing unacceptable side effects.

The drugs, DMAU and 11-MNTDC, are categorized as progestogenic androgens. These medications reduce testosterone, which reduces the number of sperm. Lowering testosterone levels often has unpleasant side effects. However, the majority of the men in the trial were willing to continue taking the drugs, indicating that the adverse effects were acceptable.

Male contraception options are currently restricted to vasectomy and condoms, and are thus extremely limited as compared to female options, said lead researcher Tamar Jacobsohn of the Contraceptive Development Program at the Eunice Kennedy Shriver National Institute of Child Health and Human Development. Development of an effective, reversible male contraceptive method will improve reproductive options for men and women, have a major impact on public health by decreasing unintended pregnancy, and allow men to have an increasingly active role in family planning.

The study consisted of two Phase 1 clinical trials with 96 healthy male participants. For 28 days, the males in each trial were given either two or four oral pills containing the active drug or a placebo. Testosterone levels fell below normal after seven days on the active drug. The testosterone levels in the placebo-taking men remained within the normal range.

The study found that 75% of men who took the active drug said they would be willing to use it in the future, compared with 46.4% of those taking a placebo. Men who took the four-pill daily dose (400 milligrams) had lower levels of testosterone than those taking the two-pill, 200-milligram dose. There was no significant difference between the two active treatment groups in satisfaction with the drug or their willingness to use it in the future or recommend it to others.

Mens positive experiences in clinical trials and high ratings of acceptability for this male pill should serve to excite the public about male birth control being potentially widely available in the coming decades, Jacobsohn said.

You might think that the colon for both women and men is pretty standard, given that we both need our digestive systems to process the food we eat, but there is one important distinction. The colon in the average female is 10 centimeters longer than a males, meaning that part of it has to lie in her pelvis, along with her bladder and reproductive organs.

A longer colon in women helps them retain fluid during pregnancy to nourish the amniotic sac. With a growing baby inside her, a woman needs much more fluid both to stay hydrated and to keep the sac filled, thereby creating the need for a longer colon compared to a mans. Gastroenterologist Dr. Robynne Chutnik describes the difference as being between a gentle horseshoe and a tangled up Slinky.

The colon in the average female is 10 centimeters longer than a males.

Because of the longer colon, the digestive process for women is much slower as theres more space to move through (sometimes taking an extra 14 hours), which makes us susceptible to digestive discomfort like bloating and constipation. Men also have more oil-producing glands, or sebum, and make about twice as much as women do. Theyre also more prone to acne and clogged pores, which are much more pronounced. When it comes to collagen, both men and women have it hard but women have it harder. Mens collagen is about 65% stronger than womens, which is one reason why men dont get cellulite. While men experience a steady decline of collagen throughout their lives, women lose theirs at a much faster pace once theyve hit menopause.

The variations dont begin and end with organs. The lumbar curve (which gives our spine its shape and curvature) is one vertebra longer in women than in men. Specifically, women have three wedged-shaped vertebrae in their lumbar region, and men have two.

The extra vertebra accommodates the shifted center of gravity during pregnancy.

This differentiation assisted early civilization when they began to walk on two feet, and womens longer lumbar curve has helped accommodate their changing center of gravity during pregnancy. One scientist for The Harvard Gazette explains, To accommodate this shifted center of gravity, womens spines have evolved to help offset the additional weight in the abdomen during pregnancy, so that the back muscles are not taxed in counter-balancing the destabilizing effects of the babys weight.

Body and facial hair have long been a significant indication of maturity and high testosterone in men. What I Learned When I Stopped Testosterone Replacement Therapy |… – Cushing’s Disease News

In 2018, I decided to stop using testosterone replacement therapy due to the side effects I was experiencing, most notably, increasingly aggressive behavior.

One concern I had at the time, though, was that ending the treatment would cause an increase in Cushings-related symptoms, because other hormones might be affected. This treatment had addressed my low testosterone levels caused by a pituitary tumor.

I do not recommend abruptly stopping treatment like I did without consulting a professional first. Although I am now seeing the benefits of my decision, it was not an easy road, and at one point, I began to question if my choice was the right one.

The first thing I noticed when I stopped taking my injections was that my mood deteriorated. Feelings of sadness led to an occasional lack of motivation. This sadness was partly due to my reluctance to stop the treatment. While I believed the medication was working, I became aware of how the side effects were affecting both me and those around me. Also, my body had grown accustomed to it over the past year, which led to more emotional imbalance.

The lows in my mood started to improve with effort, such as doing more research on ways to naturally balance testosterone levels, tinkering with areas of my life that were affected by testosterone, and being more active. Eventually, these strategies led to drastic change.

The second effect of stopping treatment was fatigue. The best way to describe it is like working outside all day in the heat. Given all of these experiences, I eventually learned that I had to give myself time. Quitting testosterone therapy wasnt a process with quick results. It helps to stay positive and motivated, and to take things one day at a time. The peaks and valleys of our journeys shouldnt discourage us from the goal of getting back to optimum health.

The decision to stop any treatment is not one to take lightly. Its always important to talk to your healthcare team before making any changes to a treatment regimen. Doctors can also help you develop a plan of action to replace a particular treatment, if needed.

Today, my energy levels are up again, Ive seen no signs of aggression in my behavior, and my overall sense of well-being has improved. Larry Wheels Gives Up Steroids, Goes On Testosterone Replacement Therapy – Generation Iron Fitness Network

Larry Wheels has released a new YouTube video announcing that he has decided to stop using steroids. The announcement comes after news of Wheels suffering a back injury. Wheels is known by many to be one of the strongest men in the world with epic lifting videos never fail to motivate. He has also been very open about his steroid use in the past. Which is why he decided to be transparent about his decision to give up on steroids.

Earlier this summer, Larry Wheels set a goal to be the lightest person to deadlift 1,000 pounds. In his training towards that goal, he recent suffered a back injury. He admits that this injury was due to rushing the training process towards his goal. The injury has also led to Wheels withdrawing from the 2022 Middle East Strongest Man competition. The injury was an eye opener for Wheels about his personal health. The fallout from the injury and path to recovery became the biggest factor in Wheels deciding to quit steroid use.

In the announcement video, Larry Wheels reflects on having used steroids for over a decade. And how his consistent use of PEDs has made it impossible for him to know what kind of lifting he is capable of off-cycle.

Much like you dont know, I dont know what I am capable of off-cycle. Now, going completely off isnt an option for me. I cannot produce testosterone naturally. So, I am going to be on testosterone replacement therapy (TRT). I tried pull cycle therapy several times when I began cycling and I was unable to recover.

Larry Wheels mentions a big caveat here. He cannot completely go off of steroid use because his body is no longer capable of producing testosterone naturally. This is a well known side effect from prolonged steroid use. So when Larry Wheels says he is quitting steroids he is still going to be implementing testosterone into his system via TRT.

Testosterone Replacement Therapy is a very different cycle compared to traditional steroid use as we know it in bodybuilding. TRT is a medical approach to simply bringing testosterone levels to normal since the body cannot do this on its own anymore. So while Larry Wheels will be using testosterone drugs it will only be to bring him to a natural state.

Larry Wheels continues to explain the process:

TRT for me would be anywhere from as low as 70 milligrams per week, as high as 175 I think 175 is a good place for me to start. If I speak with my doctor and we get my bloods done and we see that its more than whats necessary then I will decrease it, but I certainly wont increase it.

That last comment in the quote above is a key statement. Larry Wheels wants to keep his testosterone levels as minimal as possible while still staying in normal levels. He does not want to abuse TRT to bring his testosterone to more enhanced levels. After some initial bloodwork Larry Wheels will lower his t-levels as far as he can while staying healthy.

Now that Larry Wheels has realigned how he wants to handle his health will he still be aiming for a 1,000 pound deadlift? The short answer is yes but not anytime soon. Wheels comments that the deadlift goal will always be a passion of his. But he also knows that there is no way he would be able to accomplish the feat without using steroids in his current transition.

It was seem, in fact, that Larry Wheels isnt planning to stay steroid free forever. He hints at the notion of returning to steroid use after stabilizing his body, learning his off-cycle limits, and solidifying his overall health.

I am not calling quits on it, but I am putting the brakes on attempting PRs like that at the moment

Despite this hint, Larry Wheels seems to have no plans to switch back to steroid use anytime in the near future. After rushing over the past year and leading into injury, Wheels is ready to take it slow. He wants to focus on training, diet, and recovery off-cycle. Learn his new limits, learn where he can push the. And improve his overall health before taking any unnecessary risks.

Why Are People Icing Their Testicles? – VICE

Bros have tried many shocking if not borderline-questionable things in an effort to improve their health. There are those who expose their testicles to red light, and others who sun their genitals. Now, some are choosing to chill out, literally.

Health enthusiasts and biohackers are icing their testicles with ice packs and cold showers, believing that doing so helps increase testosterone levels and sperm quality, boosts energy and sex drive, and improves sleep quality.

The practice is based on the idea that excessive heat in the groin area is bad for testosterone production and sperm health.

According to Lye Diwa, a urologist based in the Philippines, testicles hang outside and separate from the rest of the body because they function better at a temperature lower than the bodys normal temperature. Warmer temperatures in the groin area tend to produce less ideal sperm, in terms of morphology and mobility.

People who ice their balls believe that cooling the testicles is a good way to boost testosterone and sperm, but nobody can say for sure that this actually works.

Although there are small studies that would attest to cold temperature and improvement in sperm quality, there are no large clinical trials that would advise icing as a means to improve sperm count, Diwa told VICE. One study even found the oppositeexposing testicles to cold temperatures decreased testosterone in 19-year-old males.

Of course, like with other biohacking practices, a lack of large clinical trials hasnt stopped people from giving this low-effort-but-big-promise technique a shot. Icing testicles is (almost) as simple as it sounds.

Some people use an ice pack, a towel with ice cubes, or a specially-designed testicle cooler, said Siim Land, an author and biohacker who personally just runs his balls under cold water from the shower. The point is simply to cool the testicles down, especially when they might be particularly hot like after a workout or sauna session.

The goal isnt to numb [your testicles]. You want to just cool them down and you can achieve that quickly, Land said, adding that all it takes is a few seconds or minutes of icing at a time to get the desired effect.

Land also advised people to wrap ice packs with cloth.

One big concern would be burns, so you probably dont want to put the ice pack directly on the skin. That can burn the skin if its too cold, he said.

Most people likely dont have the means to test for testosterone levels and

sperm quality at home, but some say icing their testicles might be responsible for a few things they can observe.

Vlad, a filmmaker who preferred to only go by his first name, goes to the sauna multiple times a week.

Because heat is harmful for sperm quality, and as my wife and I were trying to have a baby, I decided to apply ice packs [for a few seconds at a time] on my testicles during the 15 minutes I spent in the sauna. At first, it was sort of uncomfortable, but I rapidly grew fond of it, as it gives you a nice buzz and clears your mind, Vlad said.

He said that his wife, who was in her late 30s at the time, became pregnant within two months of his testicle-icing practice. Vlad admitted that he couldnt be sure if it was the ice, luck, or good health that helped them get pregnant, but said that he didnt think the ice did any harm.

Marc S., a teacher who preferred to abbreviate his last name lest his students read this story, said his job can be extremely stressful at times, so he tries to do what he can to improve his health and well-being. One thing that helps is icing his testicles.

After the icing, I feel the need to move my body. I feel much more energetic. My [eczema] has also drastically improved. Overall, I experience more energy and less stress, resulting in a much higher sex drive and the need to work out, said Marc.

Alex Mikaelsson, an investment manager, also said that icing his testicles gives him an increase in energy and libido.

I ice them before I go to the gym and, placebo or not, I feel it really helps me with increased and sustained energy for my workout. The other feeling, which was quite evident, was an increase in libido and the increased frequency of morning wood, said Mikaelsson.

Felipe Rodas, a fitness trainer and nutrition coach who also does red-light therapy and genital sunning, said icing his testicles for five to ten minutes every day, twice a day, helps him get better sleep quality.

Some skeptics say that whatever benefit people might gain from icing their testicles is minuscule in terms of what they might gain from improving their diet, exercising, and getting better sleep. Most of the testicle-icers VICE spoke to for this story said that theyre aware of this, but also think that icing their balls is a simple enough practice that doesnt take too much time or effort.

I would say icing your testes definitely works, but I would make sure that youre checking all the other boxes if you want sustained results, said Mikaelsson.

Land, the author, agreed that there could be more evidence, from randomized trials that control more variables, to support the practice. He also agreed that there are more important things people can do to improve their testosterone levels and sperm health. He still believes, however, that the practice has a positive effect, and when the risks are duly avoided, theres very little downside.

But the practice still gives others cold feet.

When I iced my balls in the sauna, I really hoped [it would work] because all the dudes were looking at me like I was a weirdo, Vlad said. It takes balls to ice them.
