Forming genitals in boys requires complex dance between genes and hormones – University of Wisconsin-Madison
The most common congenital disorders of all, especially in baby boys, are differences in a newborns sexual anatomy that is not standard female or male. In boys, they include undescended testicles, misplaced urethras, and improperly developed internal organs.
Some of these disorders may be treated with surgery and some also with hormonal supplementation. Scientists are troubled because the incidence of these disorders is increasing, especially in developed countries. While causes are unknown, researchers suspect that endocrine disruptors from environmental contaminants may be involved.

Its hard to know how something goes wrong if you dont know how it goes right, says Joan Jorgensen, a professor of comparative biosciences in the University of WisconsinMadison School of Veterinary Medicine.
So, Jorgensen, lead author Anbarasi Kothandapani, and others have set out to understand how functional reproductive systems are formed. Over several years, they studied how one gene, GLI3, coordinates the precisely timed development of the male reproductive organs. Using a mouse model, they discovered that GLI3 promotes the growth of the only cells capable of producing male hormones in a fetus. Without enough of those cells, development goes awry.
The researchers published their findings June 4 in the journal PLOS Genetics. Their works helps us understand how genes and hormones interact to develop male reproductive systems, a step toward researching why disorders in these organs have become more common and how we might prevent them.
The Jorgensen lab has long studied male and female sex differentiation in developing mice. Altogether, a male fetus must successfully have testes descend toward the scrotum, differentiate external genitals, and develop internal organs such as the prostate. Much of this process is controlled by a genetic pathway whimsically named Hedgehog after the gene was discovered in flies covered in bristles and hormones. The two most important hormones are testosterone and another named INSL3.
The GLI3 gene is part of the Hedgehog gene pathway and mutations in the gene are linked to reproductive tract disorders in humans. To study how GLI3 works, the Jorgensen lab turned to a line of mice carrying a mutation that models human mutations in the gene. In mice, the mutation is lethal before birth, so they had to study its effects in developing embryos.
The first thing we wanted to do was see if they had the same reproductive organ phenotypes as human patients, says Jorgensen. These problems include undescended testicles and disorders in urethra placement.
The mutant mice had several defects similar to humans with mutations in the gene. They experienced delays in the descent of their testicles within their abdomens And in about half of the mutants, the urethra opened at the base of the penis instead of at the tip.
Suspecting a problem with the hormone-producing cells called Leydig cells, the researchers examined the testes. Although mutant mice had typical-looking gonads, Jorgensens team saw their testes had fewer Leydig cells and these cells even grew weaker as the embryos developed.
We concluded that the cells started off okay, but they werent able to maintain their identity and they lost their function, says Jorgensen. Over time, they werent able to increase the amount of androgen synthesis needed to make sure everything develops properly.
The researchers discovered that the Hedgehog pathway, through GLI3, stimulates the formation of Leydig cells, which produce the hormones necessary to develop male sex organs. And those hormones go on to reinforce the Hedgehog pathway, which is vital for developing the prostate and penis. They concluded that most of the defects in the mutant mice could be explained by dysfunctional Leydig cells.
Treating pregnant mice with extra testosterone wasnt enough to correct all the defects, because the GLI3 gene was itself necessary, especially for producing a normal penis. But when the researchers reintroduced the non-mutant GLI3 gene into the mutant testes in culture, they were able to restore production of genes necessary for the production of testosterone.
The upshot is that researchers discovered a complex dance between genes and hormones. Any misstep in the waltz can spell disaster.
There are several environmental disruptors out there that will impact the Hedgehog pathway. When you put that together with genetic mutation, it could make some individuals more susceptible to developmental disorders than others, says Jorgensen.
This improved clarity in how the genes are supposed to function could help researchers understand how environmental toxins might be to blame for a rise in sex development disorders. That understanding is the first step toward prevention.

How ‘peer contagion’ may play into the rise of teen girls transitioning – New York Post
In the last decade, diagnosis of gender dysphoria severe discomfort in ones biological sex has exploded across the West.
Between 2016 and 2017, the number of gender surgeries for natal females in the US quadrupled; in the UK, the rates of gender dysphoria for teenage girls are up 4,400 percent over the previous decade. An ailment that typically began in early childhood, and overwhelmingly afflicted males, suddenly has a new dominant demographic: teenage girls.
In 2016, Brown University public health researcher Lisa Littman was scrolling through social media when she noticed that a group of teen girls from her small town in Rhode Island all from the same friend group had come out as transgender.
Intrigued by the statistical unlikelihood, Littman began to study the phenomenon and, in 2018, published the results. She hypothesized that transgender identification had become one more peer contagion among adolescent females. Anxiety-ridden, middle-class girls who once engaged in cutting or anorexia were now wearing binders (breast-compressing undergarments), taking testosterone and undergoing voluntary double mastectomies
I have interviewed over four dozen families whose teen daughters became caught in this current. Their stories follow a pattern: A girl never expresses any discomfort with her biological sex until puberty, when anxiety and depression descend. The girl struggles to make friends. She immerses in social media and discovers transgender gurus. Or her school holds an assembly celebrating gender journeys, or hosts a Gay-Straight Alliance club pushing gender ideology. At first, she tries out a new name and pronouns. Her school encourages her, keeping all this a secret from her parents. Then, she wants more.
So many women were once tomboys, as I was inhabiting femininity itchily, like the floral dress your mother made you wear. But tomboy doesnt exist anymore, as any teenage girl will tell you. In its place is an endless litany of gender categories, from agender to non-binary, gender fluid to trans. Imperfectly feminine girls are encouraged to consider their options.
As author J.K. Rowling recently wrote in a blog post that caused an uproar (but had many women nodding their heads): If Id found community and sympathy online that I couldnt find in my immediate environment [as a teen], I believe I could have been persuaded to turn myself into the son my father had openly said hed have preferred.
Ive spent the last year meeting transgender adults, trans-identifying teens and their parents, as well as experts in gender dysphoria, gender therapists, public school teachers, gender surgeons, endocrinologists and detransitioners young women who medically transitioned to the opposite sex, only to regret it.
While studies of detransitioners are only now being done, and numbers are hard to obtain, the detrans group on Reddit has nearly 13,000 members.
One detransitioner, Desmond, said her use of testosterone brought on severe uterine pain and necessitated a hysterectomy. No one really helped me at that point, she said. They were so easy to the pushing of hormones, of putting the thought of surgery in my head, of Yes, this will make you happier, this will make your life easier, this will get rid of your depression, this will help you become your true self rather than treating it as a way of escaping myself before really coming to terms and finding my true self again.
Our culture has both made womanhood less appealing and also greased the skids toward social and medical transition for confused teens in genuine pain. Public schools peddle gender ideology with a fervor that would make a preacher blush. Transgender social-media gurus tout the euphoria testosterone produces, and Planned Parenthood is one of the largest suppliers of testosterone and they do apprise of the risks, but on an informed consent basis. (Meaning, no therapist note is required and you leave the clinic with a very serious drug based essentially on the patients self-diagnosis.) Hundreds of plastic surgeons are happy to offer medically unnecessary double-mastectomies to girls as young as 16 without so much as a therapists note.
Teens today spend far less time in person with friends than any previous generation. Most of their social life occurs online, where feelings of inadequacy run wild and the challenge of making and keeping friends never lets up. One of the surest ways to win friends and followers is by declaring a glamorous new gender identity, and so they do. Anything to feel lonely no more.
But for many of these girls, the new identity provides no cure. The distress that catalyzed their gender journeys remains. There is no hack for adolescence. We push and scream through its blinding confusion because there is no choice but to grow up.

I was visiting with the oldest daughter the other day, in fact I visit with her often as she calls her dad daily, when she began telling me about another party and their ailments. If the answer was no, then I would respond, Well, just walk it off. In the event of a break or blood, they knew where the splints and band-aids were. If one wasnt enough, use two!
A couple of weeks ago I was in a conversation with Jeff Higgins. He made a statement regarding the fact that as men age, their hormone balance changes, and their body begins to produce more estrogen and less testosterone. I know that my testosterone level is low because after the cancer incident in 2009, the doctors have kept it at a low level. It seems that the type of cancer that I had actually feeds off of the male hormone. In addition, if my estrogen production has increased that would explain my new ability to cry at will. When I was younger, I had to pluck a nose hair in order to bring the tears. Chicks thought I was sensitive; therefore, it was a handy talent to have.
However, if this is what women are going through on a regular basis, it explains a lot! Now before you take this the wrong way, Im not making any misogynistic statements here, Im just saying that if this is the reality of the opposite gender, then it is an entirely new viewpoint for me and Im not real comfortable with it. I have never thought that I could endure pregnancy or childbirth either, but this takes everyday life to an entirely new level. I can now understand the irony of the following: Men say they wish women came with instructions. Whats the point? Have you ever seen a man read the instructions?
I liked the old me, but the new me is not the destination that I was aiming for. I really wanted to depart this world like my great-uncle Tobias, peaceful and asleep. Not like the rest of the people in the car screaming and yelling for him to get his hands back on the wheel! With this sudden heightened level of emotions, I probably fall into the more vocal group.
As this new condition tries to achieve a balance, I am undoubtedly forced to take my own advice as given to many in the past. Lets see if this works as I try to just walk it off!

A team of Spanish researchers were curious, so did a little bit of digging. They found that from a sample of 122 male COVID-19 patients, 79 percent were bald. Soon after, a report published in the Journal of Cosmetic Dermatology also found that there was a distinct lack of hair amongst COVID hospital admittees.
